Nested allele-specific PCR primers distinguish genetic groups of Uncinula necator

Article Abstract:

A nested allele-specific PCR assay, wherein eburicol 14-alpha demethylase (CYP51) point mutations were used to detect and distinguish groups I and II from group III in crude mildewed samples from vineyards, was developed. Both CYP51 and the ITS-5.8S rDNA region from Uncinula necator isolates belonging to all three genetic groups were cloned and sequenced to identify mutations that distinguish the genetic groups. It was observed that a shift from a population composed primarily of group I isolates to a population consisting primarily of group III isolates took place during the grapevine growing season.

A mutation in the 14alpha-demethylase gene of Uncinula necator that correlates with resistance to a sterol biosynthesis inhibitor

Article Abstract:

The genetic sequencing of the P-450(sub 14DM) gene of Uncinula necator showed a point mutation that is associated with the resistant phenotype (RF,>5). Polymerase chain reaction tests further showed that the gene has the same nucleotide substitution at codon 136 in 16 of the 19 isolates studied with an RF of at least 5. This gives proof that U. necator is resistant to the demethylation-inhibiting fungicide triadimenol.

PCR assays that identify the grapevine dieback fungus Eutypa lata

This paper describes a polymerase chain reaction based technique for the identification of grapevine dieback fungus Eutypa lata using nucleic acid probes derived from ribosomal DNA and polymorphic DNA fragments of the fungus.
