Nonradioactive method to study genetic profiles of natural bacterial communities by PCR-single-strand-conformation polymorphism

Article Abstract:

The combination of polymerase chain reaction (PCR) amplification and single-strand-conformation polymorphism pattern analyses of 16 rRNA genes is an effective method to study bacterial community structures in various aquatic ecosystems. This method can operate without radioactive substrates and long PCR primers possessing GC lamps. The method shows that a bacterial strain produces a characteristic band pattern that is different from the pattern produced by other strains. The method is less time consuming and laborious, and has many advantages over other methods.

Molecular analysis of microbial community structures in pristine and contaminated aquifers: field and laboratory microcosm experiments

Article Abstract:

Phylogenetic probes were used to identify alterations in aquifer microbial community structure stemming from anthropogenic perturbances using fuel contaminated and non-contaminated aquifer material sourced directly from the field as well as on aquifer materials used to establish anaerobic microcosms supplemented with toluene and/or nitrate. Alterations in aquifer microbial communities elicited by anthropogenic perturbances can be inferred from microcosm studies integrating chemical and phylogenetic probe analysis.

Increase in bacterial community diversity in subsurface aquifers receiving livestock wastewater input

Article Abstract:

The presence of rumen-related RFLP patterns in groundwater can be a sign of fecal contamination from livestock. In a study of one contaminated aquifer, Cytophaga, Flexibacter, and Bacteroides were found in greater quantities than in an uncontaminated aquifer.
