Nutrient amendments in soil DNA stable isotope probing experiments reduce the observed methanotroph diversity

Article Abstract:

The methanotroph activity and diversity in soil was examined during the consumption of [[super 13]C[H.sub.4]] with three DNA-stable isotope probing (SIP) experiments using microscosms with natural field soil water conditions, the addition of water, and the addition of mineral salts solution. DNA-SIP incubations using water-amended microcosms yielded faster incorporation of [super 13]C into active methanotrophs while avoiding the cross-feeding of [super 13]C.

Comparison of diversities and compositions of bacterial populations inhabitating natural forest soils

Article Abstract:

The diversity and composition of soil bacterial communities are compared among six Austrian natural forests, including oak-hornbeam, spruce-fir-beech, and Austrian pine forests, using terminal restriction fragment length polymorphism analysis and sequence analysis of 16 rRNA genes. The study is conducted in order to relate the bacterial community composition to the already-known characteristics of these forest soils.

mRNA-based parallel detection of active methanotroph populations by use of a diagnostic microarray

Article Abstract:

A method was developed for the mRNA-based application of microbial diagnostic mircoarrays to detect active microbial populations wherein DNA- and mRNA-based analyses of environmental samples were compared and confirmed through quantitative PCR. Results indicated that mRNA-based microarray analyses might provide additional information on the composition and functioning of microbial communities.
