Ultrastructural and immunocytochemical studies on the H2O2-producing enzyme pyranose oxidase in Phanerochaete chrysosporium grown under liquid culture conditions

Article Abstract:

Enzymatic, immunocytochemical andultrastructural studies were conducted to determine the intra- and extracelllular distribution of pyranose 2-oxidase (POD) in cultures of Phanerochaete chrysosporium K-3. The results showed that POD was preferentiallydistributed to intracellular membrane structures in electron-lucent vesicles, multivesicular bodies and in the periplasmic space of hyphae. The enzyme was also detected extracellularly. These results indicate that POD may be an important donor of hydrogen peroxide for hydrogen peroxide-dependent peroxidases during lignocellulose degradation.

Purification by immunoaffinity chromatography, characterization, and structural analysis of a thermostable pyranose oxidase from the white rot fungus Phlebiopsis gigantea

Article Abstract:

The pyranose oxidase (PROD) with a relative molecular weight of nearly 295,600 has been purified by heating, immunoaffinity chromatography and gel filtration. PROD, produced by the white rot fungus Phlebiopsis gigantea, is thermostable, yellow in color, and has a pI between pH 5.3 and 5.7. PROD is a tetrameric flavoprotein composed of ellipsoid subunits. Flavin adenine dinucleotides are covalently bound to PROD. The enzyme acts on D-glucose, L-sorbose and D-xylose, and is stable during storage.
