Rapid method for coextraction of DNA and RNA from natural environments for analysis of ribosomal DNA- and rRNA-based microbial community composition

Article Abstract:

Researchers report the first method for directly coextracting RNA and DNA from soil in order to analyze microbial communities. IT uses 16S rRNA reverse transcription-PCR and 16S ribosomal DNA-PCR.

Physiological and community responses of established grassland bacterial populations to water stress

Article Abstract:

Investigations were conducted on the effects of water stress upon the diversity and culturable activity of bacterial communities in the rhizosphere of an established upland grassland soil. Findings conclude that the imposed moisture regimen modulated the physiological status of the bacterial community and that bacterial communities in this soil are resistant to water stress.

Cytochemical colocalization and quantitation of phenotypic and genotypic characteristics in individual bacterial cells

Article Abstract:

A combination of cytochemical methods and in situ rRNA probe hybridization signals are efficient in colocalization of phenotypic and genotypic characteristics of individual bacterial cells. The technique allows the assignment of biochemical properties such as substrate-enhanced tetrazolium salt reduction and cytochrome oxidase reaction to cells detected from rRNA probes. A study of digitized images allows the determination of the relationship between rRNA signal strength and the rate of reduction of the tetrazolium salt.
