Putting it all together: building a prereplicative complex

Article Abstract:

DNA replication requires the formation of a prereplicative complex (pre-RC). It is a simple process in the viral SV40 system where the T antigen subserve the function of the pre-RC. Among eukaryotes, replication is a much more complicated reaction and requires the participation of a number of factors and the coordination of replication among many initiation sites as well as many levels of cellular control mechanisms. Some of these factors function as pre-RC.

Cyclin/Cdk-dependent initiation of DNA replication in a human cell-free system

Article Abstract:

A cell-free system initiating DNA replication under cell cycle control is presented. The system is derived from HeLa cells. The system can be used to analyze the factors that induce the initiation of DNA replication in somatic cell nuclei. Human cyclins A and E complexed to their cognate kinase Cdk2 can replace nuclear factors. Data support in vitro investigations showing a role for cyclins A and E complexed to cdk2 in G1-to-S transition in the cell cycle.
