Silencing of human polycomb target genes is associated with methylation of histone H3 Lys 27

Article Abstract:

The use of RNA expression arrays and CpG-island DNA arrays to identify and characterize human PRC2/3 target genes is reported. The results show that histone lysine methyltransferase (HKMT) Enhancer of zesta (Ez) h2 and embryonic ectoderm development (Eed), two other components of the PRC2/3 complexes, colocalize to the target promoters with Suppressor of zesta [(Su (z) 12]. The recruitment of Suz12, Ezh2 and Eed to target promoters coincides with methylation of histone H3 on Lys 27.

Histone H3 Lys 4 methylation: Caught in a bind

Article Abstract:

An attempt is made to reconcile some of the inconsistencies identified in a collection of proteins to selectively recognize di-and trimethyl H3K4, marks that are associated with actively transcribed genes. It is revealed that factors known to be involved with active transcription associate with H3K4me2/3 has important significance for understanding the functional role of this histone modification.

A silencing pathway to induce H3-K9 and H4-K20 trimethylation at constitutive heterochromatin

Article Abstract:

H3-K9 and H4-K20 trimehtylation is enriched at pericentric heterochromatin in mammals. The research data indicates a function for H4-K20 trimethylation in gene silencing and further suggest H3-K9 and H4-K20 trimethylation as important components of a repressive pathway that can index pericentric heterochromatin.
