Structure of the Rho family GTP-binding protein Cdc42 in complex with the multifunctional regulator RhoGDI

Article Abstract:

Results illustrate three-dimensional structure of the GDP-Cdc42 and RhoGDI proteins to understand structural basis for their biochemical activities. Structural data reveal that RhoGDI family of proteins regulate small GTP-binding proteins using isoprenoid moeity in these proteins.

Flipping the switch: the structural basis for signaling through the CRIB motif

Article Abstract:

This review discusses recent advances made in the cellular signaling pathways and the implications of the conserved Cdc42-and Rac-interactive binding motif (CRIB) as effectors for Cdc42 and Rac proteins. Research reveals that CRIB sequences interact with the binding domains of GTP-bound Cdc42 and Rac in mediating signaling pathways.

Activated Cdc42 sequesters c-Cbl and prevents EGF receptor degradation

Article Abstract:

Research indicates that the Ras-related protein Cdc42 blocks the binding of the c-Cbl ubiquitin ligase to the epidermal growth factor receptors, leading to accumulation of aberrant receptors and sustained epidermal growth factor-stimulated ERK activation, which results in cellular transformation. Data show that activated Cdc42 prevents the process by sequestering c-Cbl.
