Study of the organization of the genomes of Escherichia coli, Brucella melitensis and Agrobacterium tumefaciens by insertion of a unique restriction site

Article Abstract:

The introduction of the 18 bp I-SceI restriction site using Tn5Map, a derivative of Tn5, into the genomes of bacteria, which do not contain the I-SceI site, provides an effective method for genome structure determination. Pulsed-field gel electrophoresis of the Escherichia coli chromosome, containing Tn5Map, shows that the chromosome is circular in shape. Two of the Brucella melitensis chromosomes are circular. Agrobacterium tumefaciens contains a circular and a linear chromosome. The chromosomes of B. melitensis and A. tumefaciens contain many replicons.

Intra-chromosomal heterogeneity between the four 16S rRNA gene copies in the genus Veillionella: implications of phylogeny and taxonomy

Article Abstract:

Results reveal a high level of heterogeneity in sequences between copies of 16S rRNA genes in the Veillonella genome. The inter-16S rDNA-heterogeneity also exists widely among the members of the genus Veillonella. No phenotypic criteria can be identified for species differentiation. Taken together, data facor unification of V. dispar and V. parvula.

Regulation of acp1, encoding a non-aspartyl acid protease expressed during pathogenesis of Sclerotinia sclerotiorum

Article Abstract:

The gene acp1 encoding the non-aspartyl acid protease contains motifs in the promoter region, which encode binding sites that control the gene expression. Data indicate that glucose and nitrogen starvation coupled with acidification control acp1 gene expression during pathogenesis in Sclerotinia sclerotiorum.
