The RNA cleavage activity of RNA plymerase III is mediated by an essential TFIIS-like subunit and is important for transciption termination

Article Abstract:

Budding yeast RNA polymerase III (Pol III) has a small essential subunit, C11, conserved in humans. It shows much homology to TFIIS. A mutant Pol III heterocomplemented with Schizosaccharomyces pombe C11 is affected in transcription termination in vivo. Purified enzyme (Pol III delta) without C11 initiated as it should but did not deal with pause sites and in termination did not perform as it should. Pol III delta did not have the intrinsic RNA cleavage activity of Pol III in its complete form. It appears that C11 makes it possible for the enzyme to switch between RNA-elongation and RNA-cleavage modes. It seems the essential role of Pol III RNA cleavage activity is to remove kinetic barriers to the termination process. The integration of TFIIS function into a specific Pol III subunit may come from the opposite requirement of Pol II and Pol III related to transcript length and efficiency of termination.

Elongation by RNA polymerase II: The short and long of it

Article Abstract:

The process of transcript elongation by RNA polymerase II (RNAP II) is identified and the many factors that regulate the elongation stage of transcription are discussed. Elongation is highlighted as a central process that coordinates multiple stages in mRNA biogenases and maturation.
