The phosphinomethylmalate isomerase gene pmi, encoding an aconitase-like enzyme, is involved in the synthesis of phosphinothricin tripeptide in Streptomyces viridochromogenes

Article Abstract:

Researchers have discovered an enzyme in Streptomyces viridochromogenes Tu494 that is similar to aconitases from plants, bacteria, and fungi. This enzyme is involved in the biosynthesis of the antibiotic phosphinothricin tripeptide.

Biosynthetic gene cluster of the herbicide phosphinothricin tripeptide from Streptomyces viridochromogens Tu494

Article Abstract:

Uncharacterized flanking and middle regions of the antiobiotic phosphinothricin tripeptide (PTT) biosynthetic gene cluster from Streptomyces viridochromogenes Tu494 are isolated and sequenced. Four of these are identified as undescribed genes coding for proteins that are involved in uncharacterized early steps of antibiotic biosynthesis or in providing precursors of PTT biosynthesis.

The peptide synthetase gene phsA from Streptomyces viridochromogenes is not juxtaposed with other genes involved in nonribosomal biosynthesis of peptides

Article Abstract:

The Streptomyces viridochromogenes peptide synthetase gene, phsA has no surrounding peptide antibiotic encoding genes. The gene encodes a protein with 622 amino acids. The carboxy terminal of the PhsA protein is essential for the peptide synthetase activity. The gene is involved in the production of phosphinothricin tripeptide. PhsA is necessary for the activation and condensation of one amino acid. Fragments of the phsA gene cloned into pGM vectors integrate into the phsA gene or outside the phsA locus in gene disrupted mutants.
