c-Myc is an important direct target of Notch1 in T-cell acute lymphoblastic leukemia/lymphoma

Article Abstract:

A preliminary test was conducted, where the effects of [gamma]-secretase blockade on c-myc mRNA levels was analyzed in five human T-cell lymphoblastic leukemias and lymphomas (T-ALL) cell lines, that require Notch signals for growth and was further encouraged with experiments, using c-myc-dependent murine T-ALL cells. The data showed that implicate c-myc is a developmentally regulated direct downstream target of Notch1, which contributes to the growth of T-ALL cells.

Inactivation of S6 ribosomal protein gene in T lymphocytes activates a p53-dependent checkpoint response

Article Abstract:

One or two alleles of the 40S ribosomal protein S6 gene in T cells in the mouse are deleted conditionally to study the effect of impaired ribosome function in vivo. The results demonstrate the existence of a p53-dependent checkpoint mechanism that senses changes in the fidelity of the translational machinery to prevent aberrant cell division or eliminate defective T cells in vivo.

Several diaA mutant alleles were isolated using in vitro Mn-PCR mutagenesis and in vivo complementation analysis in an attempt to determine the functional mechanism of DiaA-mediated stimulation of replication initiation.
