pelle encodes a protein kinase required to establish dorsoventral polarity in the Drosophila embryo

Article Abstract:

The Drosophila tube and pelle (pll) genes are required for the intracellular signal transmission leading to the nuclear import of the dorsal protein. The pll gene was cloned and characterized. Sequence analysis showed that the pll gene codes for a protein kinase similar to the raf-mos kinases. Mutations in the kinase domain of the pll protein resulted in functional disruption. These results indicate that phosphorylation is involved in the nuclear import process.

Cellular signaling: pivoting around PDK-1

Article Abstract:

The phosphoinositide-dependent kinase-1 is a member of the ABC superfamily of protein kinases and is tightly controlled in terms of its dependency on phosphorylation for substrate conformation and subcellular localization. Research reveals that the substrate conformational regulation enables the enzyme discriminate targets leading to a selective cellular response.

Mechanisms of autoinhibition and STI-571/imatinib resistance revealed by mutagenesis of BCR-ABL

Article Abstract:

The mechanism of drug resistance of the myeloid leukemia causing Bcr-Abl fusion protein kinase, to the chemotherapy agent STI-571 is described. Mutagenesis studies show that unique variants of the protein allosterically destabilize the autoinhibited conformation of the ABL kinase, which binds TI-571.
