Requirement of monooxygenase-mediated steps for sterigmatocystin biosynthesis by Aspergillus nidulans

Article Abstract:

Several monoxygenases are required in the synthesis of sterigmatocystin in Aspergillus nidulans. They include a putative P-450 monooxygenase and a putative flavin-requiring monooxygenase. Mutant strains with defects in the genes for these enzymes accumulate averufin.

stcS, a putative P-450 monooxygenase, is required for the conversion of versicolorin A to sterigmatocystin in Aspergillus nidulans

Article Abstract:

The Aspergillus nidulans gene stcS appears to produce an enzyme that is necessary to convert versicolorin A to sterigmatocystin (ST), a carcinogenic product of metabolism. A strain of A. nidulans which had an alteration in the stcS gene could not produce ST. The gene appears to produce a cytochrome P-450 monoxygenase. The convesion of versicolorin A to ST seems to require more than one enzyme, because previous studies have shown another gene to be required in the conversion.

Aspergillus nidulans stcP encodes an O-methyltransferase that is required for sterigmatocystin biosynthesis

Article Abstract:

The Aspergillus nidulans stcP gene encodes a methyltransferase required for the biosynthesis of sterigmatocystin from demethylsterigmatocystin. The disruption of stcP gene in A. nidulans produces strains that accumulate demethylsterigmatocystin. A. nidulans produces this polyketide mycotoxin sterigmatocystin, which is a second-to-last intermediate in the aflatoxin pathway.
