Induction and monitoring of definitive and visceral endoderm differentiation of mouse ES cells

Article Abstract:

The definitive and visceral endoderm is distinguished by using a mouse embryonic stem (ES) cell line that bears the gfp and human IL2Ralpha (also known as CD25) market genes in the goosecoid (Gsc) and Sox17 loci, respectively. This cell lines allow to monitor the generation of Gsc(super +)Sox17(super +) definitive endoderm and Gsc(super -)Sox17(super +) visceral endoderm and to define culture conditions that differentially induce definitive and visceral endoderm.

Efficient differentiation of human embryonic stem cells to definitive endoderm

Article Abstract:

The potential of human embryonic stem (hES) cells to differentiate into cell types of a variety of organs has generated excitement over the possible use of hES cells in therapeutic applications. Directing hES cells to the definitive endoderm lineage is focused, as this step is a prerequisite for efficient differentiation of mature endoderm derivatives.

BMP-4 is required for hepatic specification of mouse embryonic stem cell-derived definitive endoderm

Article Abstract:

The mouse embryonic stem cells efficiently generate an endoderm progenitor population when differentiated in the presence of activin A In serum-free conditions. The findings establish a developmental pathway in embryonic stem cell differentiation cultures that leads to efficient generation of cells with an immature hepatocytic phenotype.
