Issues concerning the calculations of the Gibbs free energy and the concentration profile of the interface between an aqueous solution and a protein crystal are reported.

The protein-water phase diagram and the growth of protein crystals from aqueous solution

Article Abstract:

The phase diagram of a protein-water system was measured using a model with only two parameters that characterize the interaction between protein molecules in the liquid and in the solid phase. The resulting phase diagram was then used to analyze the optimal conditions for the growth of protein crystals. Results suggested that the preferred mechanism for the growth of protein crystals is one with liquid-liquid phase separation as a first step.

Relation between the solubility of proteins in aqueous solutions and the second virial coefficient of the solution

Article Abstract:

The solubility of the protein and the second virial coefficient of the solution are significant parameters in studies relating to developing diffraction quality protein crystals for x-ray structure determination using protein-water solutions. However, protein solubility and second virial coefficient of the solution are not independent parameters. A study was therefore conducted to derive a theoretical relation between these two parameters.
