Adrenal GRK2 upregulation mediates sympathetic overdrive in heart failure

Article Abstract:

Adrenal adrenergic receptor signaling is investigated in heart failure models to describe the failure of sympatholytic agents and lower the sympathetic nervous system (SNS) activity. Adrenal gland-specific G protein-coupled receptor kinase 2 (GRK2) inhibition has reversed [alpha.sub.2]-adrenergic receptor ([alpha.sub.2]AR) dysregulation in heart failure, thereby resulting in lowered plasma catecholamine levels, improved cardiac [beta]AR signaling and function and increased sympatholytic efficacy of [alpha.sub.2]AR agonist.

Locating heart failure

Article Abstract:

A study to show that loss of phosphodiesterase 4D (PDE4D) from the ryanodine receptor complex, a cardiac calcium channel, causes calcium leak, heart failure and arrhythmia. It is important to correct this molecular defect through therapeutic strategies.

Viagra: Now mending hearts

Article Abstract:

Sildenafil, a drug better known as Viagra, can suppress cardiac hypertrophy and improve heart function in mice exposed to chronic pressure overload. It raises cyclic GMP (cGMP) levels by inhibiting PDE5A, a phosphodiesterase that breaks down the cGMP.
