Cancer therapy using a self-replicating RNA vaccine

Article Abstract:

Naked non-infectious, self-replicating RNA may be an excellent candidate for development of new cancer vaccines, based on recent study. A gene encoding an RNA replicase polyprotein from the Semliki forest virus, in combination with a model antigen, was used in a successful effort to make a nucleic acid vaccine self-replicating.

Alphavirus-based DNA vaccine breaks immunological tolerance by activating innate antiviral pathways

Article Abstract:

Results show that vaccines comprised of naked DNA encoding an alphavirus replicon and the self/tumor antigen break tolerance and confer immunity to melanoma. Data indicte that the vaccine mediates double-stranded RNA production, which is crucial for vaccine function.

Interferon-producing killer dendritic cells provide a link between innate and adaptive immunity

Article Abstract:

A third dendritic cells (DCs) lineage termed interferon-producing killer DCs (IKDCs) are described. IKDCs are capable of killing typical natural killer (NK) target cells using NK-activating receptors upon stimulation with CpG oligodeoxynucleotides.
