Chronic inhibition of cyclic GMP phosphodiesterase 5A prevents and reverses cardiac hypertrophy

Article Abstract:

The chamber and myocyte hypertrophy is suppressed on blocking the intrinsic catabolism of cGMP with an oral phosphodiesterase -5A (PDE5A) inhibitor, which also improves in vivo heart function in mice exposed to chronic pressure overload induced by transverse aortic constriction. The inhibitor reverses pre-established hypertrophy induced by pressure load while restoring chamber function to normal.

PKC-alpha regulates cardiac contractility and propensity toward heart failure

Article Abstract:

The protein kinase C (PKC)-alpha is identified as a fundamental regulator of cardiac contractility and Ca (+2 super) handling in myocytes. Results of the study suggest PKC-alpha functions as a nodal integrator of cardiac contractility by sensing intracellular Ca (+2 super) and signal transduction events, which can profoundly affect propensity toward heart failure.

Calcium handler mishandles heart

A stress-response protein kinase C-alpha (PKC-alpha) is linked to abnormalities in calcium handling and worsening of heart failure. The inner workings of this key regulator are discussed.
