Lethality-based selection of recombinant genes in mammalian cells: application to identifying tumor antigens

Article Abstract:

A new and efficeint method is developed for the identification of genes that encode CTL target epitopes. The strategy in which a representative tumor cDNA library in a modified vaccinia virus expression vector is constructed is applied to selection of a gene that encodes a cytotoxic T-cell target antigen common to several independently derived tumors.

Virus smuggling, tax evasion and tumor assassination

Article Abstract:

A new approach is proposed in order to achieve delivery of viral vectors to systemically distributed metastases in immunocompetent hosts that smuggles these vectors through the circulation and into the tumor bed. This is done by loading the tumor-killing virus into a cell type that homes to tumors, thereby evading the antiviral immune response.

Bystander elimination of antigen loss variants in established tumors

Article Abstract:

Cancers express antigens that are targets for specific cytotoxic T lymphocytes (CTLs). Studies highlight the general importance of targeting the tumor stroma to prevent the escape of variant cancer cells.
