Novel tools for localizing ion channels in living cells

Article Abstract:

Several techniques can be utilized to analyze ion channel distributions in cells and tissues. The concept of selective ion flow through potassium channels has been utilized successfully to characterize a crystallization technique to determine function on the surface of oocytes. Moreover, a number of functional and structural tools are being developed for microscopic and nanoscopic localization studies. These include atomic-force microscopy, crystallization, polarized channel distribution and antibody labeling.

Ca2+ channel block and inactivation: common molecular determinants

Article Abstract:

Research on cloned Ca2+ channels has provided scientists with insights into the molecular structure of receptor sites for Ca2+ antagonists such as 1,4-dihydropyridines (DHPs), benzothiazepines (BTZs) and phenylalkylamines (PAAs). Fort example, the transfer of key amino acids involved in sensitivity to DHPs, PAAs and BTZs results in the transfer of the high-affinity binding sites and the whole complex of properties characterizing the drug's interaction with the native donor L-type channels.

A small K+ channel looms large

Article Abstract:

Pharmacologists are closely studying the mechanisms involved in ion channels, particularly K+ channels, to understand the drug specificity and action. Pharmacologic agents such as class III antiarrythmics which block K+ channels are being studied to determine their effects on cellular systems. Pharmacologists are also analyzing the mechanism behind voltage-dependent K+ channel regulation and its impact on heterologous systems.
