A multivalent PDZ-domain protein assembles signalling complexes in a G-protein-coupled cascade

Article Abstract:

The response by cells to external signals is mediated by plasma-membrane receptors, which are able to maintain the specificity of signalling. This may be due to the organization of different signalling cascades into physically and functionally distinct signalling units. The Drosophila phototransduction cascade has similarities with other signalling cascades, and its inaD gene encodes five PDZ domain proteins. Studies show the presence of a multivalent adapter protein linking the signalling components of a cascade, and show that PDZ domains are fundemental to transduction complex assembly.

Assembly of the Drosophila phototransduction cascade into a signalling complex shapes elementary responses

Article Abstract:

The InaD protein in Drospophila photoreceptors, comprises five PDZ domains. Single-photon responses in several photoreceptor mutant backgrounds have been studied and it is shown that the INaD macromolecular complex is a signalling unit underlying elementary responses. Localized activity of the signalling unit is shown to promote reliable single-photon responses, along with rapid activation and feedback regulation.

Regulation of PLC-mediated signalling in vivo by CDP-diacylglycerol synthase

Article Abstract:

Phospholipase C (PLC)-mediated signalling, which is a special type of intracellular signal transduction in eukaryotic cells, is regulated in vivo by the enzyme CDP-diacylglycerol synthase (CDS). The regulation process is performed by the photoreceptor cell-specific isoform of CDS. This isoform mediates the regulation by using the G-protein-coupled signalling.
