Early events in the thymus affect the balance of effectors and regulatory T cells

Article Abstract:

A study questions the completeness of the interpretation that critical balance between effector and regulatory mechanism is highlighted by serious immunopathologies attributable to mutations in Foxp3, a transcription factor required for a major subset of regulatory T [T.sub.a] cells in cellular immunology. It is shown that without any obvious effect on T-cell receptor-mediated selection, the normal differentiation of mouse (gamma)(delta) TR cells into potent cytolytic and interferon-gamma-secreting effector cells is switched towards an aggregate regulatory phenotype.

Deficiency of presenilin-1 inhibits the normal cleavage of amyloid precursor protein

Article Abstract:

The presenilin-1 (PS1) gene is involved in gamma-secretase-mediated proteolytic cleavage of the C-terminal transmembrane fragments of the amyloid precursor protein following their generation by alpha- and beta-secretase(s). It is not likely that PS1 is the elusive gamma-secretase, or one of the gamma-secretases, as there is a lack of sequence identity or structural homology between PS1 and any proteinase domain known. It appears that clinical mutations of PS1 lead to a gain of the function of PS1 and are not a haplotype insufficiency.

Photoactivated gamma-secretase inhibitors directed to the active site covalently label presenilin 1

Article Abstract:

Photoaffinity labelling of presenilin 1 and presenilin 2 by strong gamma-secretase inhibitors designed to act as transition state analogue inhibitors directed to the active site of an aspartyl protease is discussed. It is suggested that presenilin 1 and presenilin 2 could contain the active site of gamma-secretase.
