Effects of NF-kappa-B1/p105 processing of the interaction between the HTLV-1 transactivator Tax and the proteasome

Article Abstract:

The viral Tax protein, encoded by human T-cell leukaemia virus (HTLV)-I, binds to two subunits of the 20S proteasome, HsN3 and HC9. The stimulation of the processing of the precursor protein NF-kappa-B1/p105 to p50 occurs due to strengthening of p105-HC9 interaction by Tax. The interaction of Tax with HsN3 modifies the proteasome activity during the p105 processing. Tax may induce a direct loading of p105 onto the 20S proteasome or favor binding of ubiquitinated p105 to the 26S proteasome.

Mechanism of DNA-binding enhancement by the human T-cell leukaemia virus transactivator Tax

Article Abstract:

The human T-cell leukemia virus transactivator Tax interacts with the basic segment of proteins' bZIP elements to both increase stability and DNA affinity. Two possible mechanisms are most likely involved: either the orientation of the basic-spacer segment helices, making DNA binding easier by reducing the entropy or stabilization of a single basic-spacer helical structure in the unfolded region.
