Insights into Wnt binding and signalling from the structures of two frizzled cysteine-rich domains

Article Abstract:

Research describing cysteine rich domains in proteins is presented. In particular Wnt signalling proteins are investigated with respect to the amino terminus of Frizzled proteins.

A new member of the frizzled family from Drosophila functions as a Wingless receptor

Article Abstract:

Addition of the Wingless protein to cultured Drosophila cells containing a frizzled Drosophila gene, Dfz2, results in elevation of the Armadillo protein. In addition, human cells that contain Dfz2 are also influenced by the Wingless protein. This indicates that Dfz2, in particular, is a Wingless receptor. It also implies that Wnt signalling molecules bind to Frizzled proteins in general.

A new secreted protein than binds to Wnt proteins and inhibits their activities

Article Abstract:

The Wnt proteins are a large group of extracellular signalling molecules required in a wide range of normal and pathological developmental process in the animal kingdom. The Wnt-inhibitory factor-1 (WIF-1) secreted protein is seen in fish, amphibia and mammals, and expressed during Xenopus and zebrafish development. Xenopus embryos were used to show that WIF-1 overexpession has an affect on somitogenesis, indicating that Wnt proteins interact with diverse extracellular inhibitors.
