Site-specific chromosomal mutagenesis using the TargeTron(super TM) Gene Knockout System

Article Abstract:

The TargeTron(super TM) Gene Knockout System is a novel prokaryotic functional genomics tool based on the Ll.LtrB group II intron from Lactococcus lactis. The site-specific nature of TargeTron insertion allows step-wise mutagenesis into the N-terminal essential region to estimate the maximum truncation the RNase E gene could tolerate and retain cell viability.

Article Abstract:

Mutants are created with the change in the enzyme's component amino acids that can be used to synthesize pharmaceutical building blocks. As a result, the emphasis in enzyme engineering shifted from using rational design to directed-evolution tools that rely on random mutagenesis, which creates enormous number of mutant enzymes.

Acute mutation of reinoblastoma gene function is sufficient for cell cycle re-entry

Article Abstract:

Research shows that acute loss of retinoblastoma gene in primary quiescent cells lets them into cell cycle, which elicit phenotypic expressions diffeent from germline loss of retinoblastoma gene function. However, acute loss of retinoblastoma gene in senescence cells reverses cellular senescence programme.
