Structural basis for 5'-end -specific recognition of guide RNA by the A. fulgidus Piwi protein

Article Abstract:

The crystal structure of Archaeoglobus fulgidus Piwi protein is bound to double-stranded RNA, thereby identifying the binding pocket for guide-strand 5'-end recognition and providing insight into guide-strand-mediated messenger RNA target recognition. The structure of the Piwi-RNA complex, provide direct support for the 5' region of the guide RNA serving as a nucleation site for pairing with target mRNA and for a fixed distance separating the RISC-mediated mRNA cleavage site from the anchored 5' end of the guide RNA.

Crystal structure of a human membrane protein involved in cysteinyl leukotriene biosynthesis

Article Abstract:

X-ray crystallography is used to finding the atomic structure of human cysteinyl leukotrienes synthase ((LT)[C.sub.4]S) in a complex with glutathione. The results have provided a structural basis for the development of LT[C.sub.4]S inhibitors for a proinflammatory pathway mediated by three cysteinyl leukotriene ligands whose stability and potency are different and by multiple cysteinyl leukotriene receptors whose functions are non-redundant.

Repetitive shuttling of a motor protein on DNA

Article Abstract:

The repetitive shuttling is likely to be caused by a blockade-induced protein conformational change that enhances DNA affinity for protein's secondary DNA binding site, resulting in a transient DNA loop. Repetitive shuttling is also observed on ssDNA bounded by a stalled replication fork and an Okazaki fragment analogue, and the presence of Rep delayed formation of a filament of recombination protein RecA on ssDNA.
