CTCF mediates methylation-sensitive enhancer-blocking activity at the H19/Igf2 locus

Article Abstract:

It has been possible to undertake research supporting the view that Igf2 imprinting needs a chromatin boundary element that is situated between the gene and enhancers 3' of the imprinted-control region. It is likely that the CTCF protein mediates this function in vivo.

Location of enhancers is essential for the imprinting of H19 and Igf2 genes

Article Abstract:

The H19 and Igf2 genes are closely linked, and expressed in endodermal and mesodermal tissues. A proposed model explains the reciprocal imprinting of the genes, with expression ruled by competition between their promoters. A further set of enhancers could be predicted to relive the competition and this was tested by generating mice with a duplication of the enhancers.

The temporal requirement for endothelin receptor-B signalling during neural crest development

Article Abstract:

Endothelin receptor B (EDNRB) is a G-protein-coupled receptor required for development of melanocytes and enteric neurons. The tetracycline-inducible system was exploited to generate strains of mice where the endogenous Ednrb locus is controlled by tetracycline-dependent transactivators tTa or rtTA. It was determined that EDNRB is needed during a restricted period of neural crest development between embryonic days 10 and 12.5.
