Genetic contributions to understanding polyketide synthases
Article Abstract:
There have been significant advances in the understanding of the structure and mechanisms of polyketide synthases (PKSs), using chemical and biochemical approaches. An historical view of the understanding of PKSs obtained through genetic approaches, such as sequence analysis, mutagenesis and gene expression, is provided, and is mainly concerned with the streptomycetes and their close relatives among the actinomycetes.

Harnessing the biosynthetic potential of modular polyketide synthases
Article Abstract:
The discovery of modular polyketide synthases (PKSs), found in Gram-positive and Gram-negative bacteria, has resulted in new horizons for the engineered biosynthesis of complex natural product-like molecules. A review of insights into the structure and properties of modular PKSs over the past 10 years is presented, and the currently available methods of using modular PKSs to synthesize novel biomolecules are assessed.

Manipulation of modular polyketide synthases
Article Abstract:
Polyketides are synthesized by bacteria, fungi and plants via condensation of carboxylic acids, and are widely used in human and veterinary medicine, animal nutrition and agriculture. They are grouped into aromatic and complex classes, and the complex class is structurally more diverse. Biosynthesis of the complex polyketides is discussed, and an overview of genetic changes in PKS genes is provided.
