Mechanisms of developmental control of transcription in the murine alpha- and beta-globin loci

Article Abstract:

Messenger RNA transcription and expression of the mouse alpha- and beta-globin loci in development have been characterized. All seven murine globin genes are transcribed in primitive erythropoiesis, but transcription of three of them is restricted to the primitive erythroid lineage. There is evidence of EKLF activity in embryonic red cells. It may be that there is multigene expression in the beta locus takes place though alternating single-gene transcription although coinitiation may be part of the alpha locus. It seems that the multigene expression mechanism of alternation transcription is evolutionarily conserved from mouse to human beta globin loci. Timing of activation of the adult genes is not the same, however, so important fundamental differences likely exist in globin gene switching.

The active spatial organization of the beta-globin locus requires the transcription factor EKLF

Article Abstract:

Three-dimensional organization of a gene locus is important for its regulation, as recently demonstrated for the beta-globin locus. It is reported that erythroid Kruppel-like transcription factor (EKLF), an erythroid transcription factor required for adult beta-globin gene transcription is also required for active chromatin Hub (ACH).
