Use of the KlADH4 promoter for ethanol-dependent production of recombinant human serum albumin in Kluyveromyces lactis

Article Abstract:

The ethanol-driven expression of bacterial beta-glucuronidase and human serum albumin (HSA) genes in Kluyveromyces lactis through the KlADH4 promoter has been studied. The KlADH4 promoter is a gene found in K. lactis which encodes a mitochondrial alcohol dehydrogenase activity induced by ethanol. K. lactis is an aerobic yeast which uses lactose as its sole carbon source. Results show an increase in the extracellular production of recombinant HSA with integrative and replicative vectors. It was also observed that there was no limitation in the KlADH4 trans-acting factors found in the cells.

Gene cloning, DNA sequencing, and expression of thermostable beta-mannanase from Bacillus stearothermophilus

Article Abstract:

Research was conducted to examine the DNA sequencing, gene cloning and the expression of thermostable beta-mannanase from Bacillus stearothermophilus. The detection of recombinant beta mannanase activity was done based on the clearing of halos surrounding the colonies of Escherichia coli grown on a dye-labelled substrate. The high level of expression of recombinant mannanase in addition to its thermostability indicate its possible commercial use in biobleaching.

Expression and secretion of a thermostable bacterial xylanase in Kluyveromyces lactis

Article Abstract:

The xylanase gene xynA was isolated from anaerobic bacterium Dictyoglomus thermophilum Rt46B.1 and has the potential to be used in the bleaching of wood chips. The xynA gene was expressed as an extracellular protein by fusing it with the secretion signal of the yeast Kluyveromyces lactis. The xynA gene was expressed as a 35-KDa protein that made up 90% of all the extracellular proteins at a concentration of 130 microgram per milliliter in shake-flash cultures.
