Conformational variations in an infectious protein determine prion strain differences

Article Abstract:

A high efficiency protocol for infecting yeast with the [PSI(super +)] prion using amyloids composed of a recombinant Sup35 fragment (Sup-NM) is reported. The reports establish that Sup-NM adopts an infectious conformation before entering the cell - fulfilling a key prediction of the prion hypothesis- and directly demonstrate that differences in the conformation of the infectious protein determine prion strain variation.

Generation of prion transmission barriers by mutational control of amyloid conformations

Article Abstract:

Point mutations in the protein determinant of yeast prion alter conformational states of amyloid leads to the generation of prion transmission barriers but without affecting amyloid formation. Data indicate that these mutations change amyloid seeding specificity and the transmission barriers selectively disfavour subsets of prion strains.

Conformational diversity in a yeast prion dictates its seeding specificity

Research is presented concerning the use of a yeast prion to investigate the ability of prions, made up of the same polypeptide, to produce different phenotypes when propagated within hosts which are genetically identical.
