Modulating substrate choice: The SspB adaptor delivers a regulator of the extracytoplasmic-stress response to the AAA+ protease ClpXP for degradation

Article Abstract:

Proteins captured by ClpXP(super trap) in sspB+ but not sspB(super -) strains are examined to identify new sspB(super -)regulated substrates. RseA(super 1-108), a fragment of a transmembrane protein that regulates the extracytoplasmic-stress response, fits this criterion and in response to stress. RseA is cleaved on each side of the membrane and is released as a cytoplasmic fragment that remains bound in an inhibitory complex with the sigma(super E) transcription factor.

C1pS modulates but is not essential for bacterial N-end rule degradation

Article Abstract:

A genetic screen for N-terminal sequences that direct degradation of a conditionally toxic Escherichia coli protein was developed to explore the bacterial degradation of N-end rule substrates by the [AAA.sup.+] protease C1pAP. The results indicate that C1pA has an intrinsic ability to recognize and degrade N-end rule substrates whereas C1pS modulates substrate recognition and turnover.

Latent ClpX-recognition signals ensure LexA destruction after DNA damage

Article Abstract:

Research has been conducted on DNA-damage response gene upregulation when LexA repressor undergoes autocatalytic cleavage. The authors demonstrate the LexA fragments are the substrates for ClpXP protease.
