Pathophysiologically based treatment interventions in schizophrenia

Article Abstract:

The contributions of disturbances in glutamate, dopamine and GABA neurotransmission to the pathophysiology of working memory impairments in schizophrenia are considered. A cascade of molecular events that may link these disturbances are suggested and an argument that the molecular alterations most proximal to the pathophysiology of prefrontal dysfunction offer the most promise as targets for new drug development is presented.

Mutations in antiquitin in individuals with pyridoxine-dependent seizures

Article Abstract:

Pyridoxal 5'-phosphate (PLP) is an essential cofactor for many enzymatic reactions, including those involved in neurotransmitter metabolism. Individuals with pyridoxine-dependent seizures (PDS) need an unusually large daily intake of the PLP precursor pyridoxine to prevent fits, however, while it is known that PDS maps to 5q31, the mechanism of pyridoxine dependency in PDS is identified.
